Dunston take a 1-0 lead to Derbyshire after Andrew Bulford scored the only goal in the first leg.
The winners of the tie will play either West Auckland or Herne Bay in the showpiece final at Wembley in May.
Club captain Ben Cattanach told Sky Tyne and Wear: “It would mean everything. When you are growing up you watch the FA cup final on TV.
“You always dreamed about it, and if your not going to make it as a professional this is the next best thing.”
