The Eagles started brightly and were 22-11 up by the end of the first quarter. They were led by Charles Smith who scored 25 points on the night.
Joe Chapman contributed 16 points and Paul Gause hit 15 before fouling out in the third quarter.
The game was level with 44 seconds left when Arnas Kazlauskas hit a three-pointer for the Wolves to set up a dramatic ending.
Smith restored a lead of two for the home side but Sherrad Prezzie-Blue cancelled that out with a two-pointer and another from the line.
The Eagles called a timeout with just 12 seconds left and from the following play Andrew Thompson scrambled home the winning points.
Jubilant player/head coach Fabulous Flournoy told Sky Tyne and Wear: “It is a huge result but more importantly it is a statement made going into the playoffs.
“We will remember this for a long time. I want this moment to last. Happy Easter because the Easter bunny came to the Newcastle Eagles.”
Elsewhere Plymouth Raiders came from behind to win 100-93 at Milton Keynes Lions and move above Worcester in the league.
Sheffield Sharks cemented their place in the playoffs beating Durham Wildcats 79-73.
Newcastle Eagles are now six points clear at the top and ended Worcester’s chance of winning the title because of their superior record head-to-head.
